General Description of the role :

We seek a highly motivated and independent Senior Scientist to join our iPSC team in the Cell Therapy group of Janssen Biotherapeutics. The individual will be responsible for optimization of the platform for the rapid and robust generation of genetically engineered iPSC lines to support multiple internal programs. Key responsibilities include but are not limited to: serving as a subject matter expert, providing research support for the growth, maintenance and optimization of iPSC cultures and their differentiation into various immunological cell types. The individual will interact with various teams focused on engineered cell therapy platform development. The successful candidate will utilize a full suite of advanced technologies to develop best-in-class, off-the-shelf cell therapy drug candidates.

Primary responsibilities:

  • Independently design and execute experiments to optimize protocols for iPSC differentiation to therapeutic immune cells including gamma delta T cells, NK cells, and more

  • Extensive phenotypic characterization of lymphoid cells using multicolor flow cytometry.

  • Functional characterization of iPSC-derived lymphocytes, gamma delta T and NK cell assays such as proliferation, cytotoxicity, cytokine release, and ADCC assays

  • Familiarity with genetic modification of iPSCs and/or immune cells to improve safety and potency of engineered cell therapies

  • Viral and non-viral delivery methods, ELISA, qRT-PCR, cell-based potency assays

  • Coordinating experiments with other scientists across functional groups, compile data and present results and stay current on relevant primary literature

  • Partner with Manufacturing to transfer research process and adapt as needed.

  • Effectively manage multiple projects while adhering to aggressive timelines

  • Accurately record and document results in a timely fashion

  • Foster a collaborative, creative, and rigorous culture of scientific discovery

Qualifications

  • PhD degree with 7+ years research experience in immunology, oncology or related field and with experience in iPSC biology and differentiation protocols (preferably to immune lineages) is required

  • Extensive experience in platform technology development with specific knowledge in iPSC handling, selection and characterization is required

  • Preferred: Prior experience in research and development of therapeutic modalities for immuno-oncology applications

  • Experience with gene editing methodologies such as CRISPR, TALEN, ZFNs is required

  • Demonstrated ability in managing projects in a highly matrixed environment to lead cross-functional activities is required

  • Strong experience in molecular and cellular biology, and assay development with exposure to cutting-edge cell engineering techniques is preferred

  • Working knowledge of protein biochemistry and basic gene editing technologies, basic flow cytometry or cell imaging technologies and immune cell assays is required

  • Extensive experience with mammalian cell culture at small and large scales is required

  • Excellent communication (both written and oral), time management, record keeping and data analysis and outstanding collaborative skills
