The Senior Associate Scientist, Gene Therapy Immune Mitigation will work within the Gene Therapy group and dedicate to the development of Janssen’s gene delivery platforms. The candidate will play a critical role in mitigating immune reactions against the capsid and/or the transgene in the context of AAV-mediated gene therapy.

In this role, you will establish and perform in vitro, ex vivo, and in vivo immunological assays to characterize innate and adaptive immune responses to AAV vectors. You will also assess the impact of novel immune-mitigation strategies on the levels of transduction and transgene expression after systemic and/or local AAV vector injection. The Senior Research Associate will be a member of a highly cross disciplinary and collaborative team.

Key Responsibilities:

  • Develop and perform assays, such as ELISA, MSD, Luminex, immunofluorescent staining, confocal imaging, multicolor flow cytometry, and ELISpot to analyze innate and adaptive immune reactions triggered by AAV transduction.

  • Plan, prepare, and perform experiments on the bench to characterize transgene expression and transduction efficiency in various biological samples using qPCR, ddPCR, RT-qPCR, flow cytometry and/or immunohistochemistry.

  • Provide hand-on support for execution of in vivo rodent studies to characterize immune responses against AAV vectors after systemic or ocular delivery, as needed.

  • Provide hands-on support for the processing of human or animal samples for ex vivo analysis and/or histology, as needed.

  • Analyze results, prepare technical reports, and accurately document work in an electronic lab notebook (ELN). Prepare Powerpoint presentations of results. Communicate results and research progress during team meetings as needed.

  • Maintain inventories and/or place orders of reagents and materials needed for the assigned projects for timely execution of related lab work.

  • Assist with the maintenance, calibration, and management of laboratory equipment.

Qualifications

Education:

  • A minimum of a Bachelor’s degree in a relevant field is required

  • Master’s degree preferred

Experience And Skills:

Required:

  • A minimum of 2 years of hands-on proven experience with immunology or virology is required.

  • Direct experience and skills in flow cytometry (multicolor and sorting), cytokine assays (ELISA/MSD, Luminex, ELISpot), and immunofluorescent staining and imaging are required.

  • Experience with mammalian cell culture and processing/handling of primary cells is required.

  • Excellent work planning, organization, record keeping, and attention to detail.

  • Ability to work in a hands-on laboratory setting after initial training, develop new techniques, manage multiple tasks, and demands, enforce instructions, and follow company policies.

  • Desire to learn new skills, take on new ideas, branch out into new territory and collaborate with scientists all over the organization.

  • Works with colleagues as a great teammate.

Preferred:

  • Experience with AAV gene therapy is a plus.

  • Familiarity with ELN is beneficial.
