ABOUT THE RESEARCH INSTITUTE:
The HSS Research Institute is dedicated to research on identifying mechanisms underlying bone biology and rheumatic conditions, and developing effective approaches for prevention, diagnosis, and treatment. With a focus on tissue repair, improving surgical outcomes, autoimmunity and inflammation, genomics, new treatments, and precision medicine, the impact of our discoveries determine future directions of musculoskeletal research to best improve clinical outcomes, while serving the community in an environment that aspires to scientific excellence.
JOB SUMMARY:
The Rogatsky Lab is a basic science research lab of 5-8 people composed of graduate students, postdocs, and occasionally undergraduate interns, with an extremely collaborative, diverse and inclusive environment. There are three main projects in the lab: 1) The role of transcriptional coregulator GRIP1 in neuroinflammation; 2) RNA Pol II pausing in macrophage-driven inflammation; 3) transcriptional programming of the homeostatic (M2-like) macrophages and the role of GRIP1 in this process.
The Postbac will learn the techniques that are shared between all three of projects, which are assessment of gene expression in human and mouse macrophages or microglia by RT-qPCR and RNAseq; isolation/purification and culture of primary human and mouse macrophages; assessment of transcription factor occupancy and function and chromatin state by ChIP-qPCR and ChIPseq/CUT&RUN, ATACseq; FACS analysis and mouse genotyping and husbandry. The primary project for the Postbac is RNA Pol II pausing in macrophage-driven inflammation. Key aspects of the project are working with primary mouse and human macrophages as well as frozen synovial cell suspensions from patients and evaluating gene expression and epigenomic make-up following differential treatments with inflammatory and anti-inflammatory triggers. In vivo work will include using mouse models of arthritis and other inflammatory conditions to assess efficacy of select compounds as anti-inflammatories.
Qualifications
Education:
BS in Biology/Chemistry, Basic experience in the research lab, RNA/DNA work, some experience working with mice, cell culture
Experience:
Prefer experience working in a research lab.
ADDITIONAL SKILLS:
Exceptional organizational skills
Strong written and communication skills
Proficient use of computers and software, including Microsoft Excel and Word
Self-motivated and able to work independently and as part of a team
