Will help maintain mouse lines by setting up breeding cages and separating and genotyping offspring. Will set up timed mattings and perform dissections to collect tissues for imaging and molecular studies of in vivo placental development. Will use sterile tissue culture methods to maintain human placenta stem cells, generate knock-out and knock-in clonal lines and analyze the differentiation of stem cells using 3D culturing techniques to generate key cell types of the placenta in vitro. Will use molecular techniques such as genotyping, endpoint PCR, ELISA, western blot and qRT-PCR as well as immunofluorescence microscopy and live cell imaging to characterize differentiated cell types. Will prepare samples for genome-wide analysis using next-generation sequencing. Will maintain a laboratory notebook and update shared laboratory protocols and ensure adherence to regulatory policies of the University. Responsibilities: 15% - Oversee the use and maintenance of laboratory equipment, training personnel/volunteers, and stocking/ordering of consumable lab supplies. 25% - Oversee multiple tissue culture lines using sterile techniques. Perform lentiviral transduction and transient transfections, isolate clonal cell lines and extract DNA and RNA to identify lines of interest. 20% - Perform immunostaining and fluorescence microscopy of formaldehyde fixed tissues and cells. Optimize antibody staining conditions and acquire multicolor high resolution images using a fluorescence microscope. Maintain a repository of collected images and associated metadata. 20% - Help oversee a mouse colony by setting up breeding cages, weaning, genotyping, and maintaining records in a centralized database. Timed matings and dissection will also be used to assess the embryonic development of the mouse placenta from embryonic day 6.5 to 14.5. 10% - Perform general molecular biology assays including genotyping, endpoint PCR, qRT-PCR, gel electrophoresis, western blotting, cloning and DNA plasmid preparation. 10% - Support detailed records of all experiments and procedures in laboratory notebooks and in online databases and shared folders. Maintain and update shared laboratory protocols. Qualifications Required Qualifications: BS/BA and 1 years of related experience or equivalent combination a combination of education and work experience to equal five years. Knowledge of molecular biology techniques and molecular genetics is required. Applicant must also have excellent organizational and communication skills and be highly self-motivated.
