Position Summary

The overarching goal of the Center for Catalysis in Biomimetic Confinement (CCBC) is to understand the means by which nature spatially organizes catalysis across scales using compartmentalization in bacteria. The team of investigators from Michigan State University, Argonne National Laboratory and Lawrence Berkeley National Laboratory will work together to acquire a fundamental understanding of how multi-step reaction pathways are confined within and optimized by selectively permeable protein-based shells ofBacterial Microcompartments (https://www.nature.com/articles/s41467-021-24126-4) (BMCs). The CCBC will apply this knowledge to establish BMC shell proteins as building blocks that can be used for confined, hierarchically-ordered biological and synthetic multi-step catalysis for reactions with long-range goals that involve addressing global challenges related to energy and the environment. The Parent lab (https://kparentlab.natsci.msu.edu/) traditionally studies complex protein assemblies such as bacteriophage and giant viruses, and is applying their expertise in single particle cryo-EM to understand BMC shell assemblies.

Department Statement

Cryo-EM facilities available at MSU

Michigan State University houses a number of state of the art core facilities under the umbrella of the “Research Technology Support Facility” including a modern cryo-EM suite (https://cryo-em.natsci.msu.edu/about/index.aspx). This includes a Talos Arctica outfitted with an autoloader, Falcon4i direct detector and a Selectris energy filter. A Titan Krios G4 with cold FEG, Falcon 4i and SelectrisX energy filter equipped with a micro-ED package will be installed this summer. In addition, we will be installing a Helios Hydra FIB-SEM instrument this summer as well. Cryo-EM or other structural biology method experience is helpful, but not necessary. Training opportunities for all aspects of grid preparation, sample imaging, and post imaging analysis are available.
