Case Western Reserve University Research Technician 1 in Cleveland, Ohio
Working under close supervision, the Research Technician 1 will perform routine molecular biology and protein biochemistry techniques to support various research projects. Responsibilities include cloning, protein expression and purification, nucleosome preparation, and DNA preparation. This role will aid research into genetic and biochemical pathways by providing critical support for experiments and maintaining laboratory equipment. No animal work is involved. This is a part-time position.
ESSENTIAL FUNCTIONS
- Set up specified supplies and equipment according to detailed specifications. Prepare and set up equipment necessary for molecular cloning and mutagenesis, including PCR machines and gel electrophoresis units, to ensure accurate and efficient gene cloning processes. Regularly calibrate and set up the AKTA system for protein purification, ensuring all necessary components are operational and ready for culturing and purification tasks. (30%)
- Prepare solutions and reagents essential for PCR, protein purification, nucleosome assembly, loading dyes, and reagents for structural studies. Mix and maintain accurate concentrations of solutions required for bacterial culture and protein purification, including buffers for Ni-NTA and ion-exchange columns. (20%)
- Maintain records. Keep detailed records of cloning and gene manipulation procedures, including primers, plasmid maps, transformation efficiencies, and sequencing results. Document protein expression and purification runs, including detailed notes on purification yields, buffer conditions, and adjustments made during AKTA system operation. (20%)
- May perform inventory control, ordering, and recording of experiment. Manage inventory of lab essentials such as agar, antibiotics, Ni-NTA resin, and other consumables. Ensure essential reagents and lab supplies are available to support daily operations. Order and track stock of critical lab items, including specialized enzymes for cloning and nucleosome preparation, to maintain workflow continuity. (10%)
- May operate single controls on equipment and make light repairs. Operate basic lab equipment, including gel documentation systems and centrifuges, and perform routine maintenance and troubleshooting as needed. Ensure the AKTA system is clean and functional, performing light maintenance on pumps and tubing to support consistent protein purification tasks. (10%)
Perform housekeeping tasks. Keep work areas organized and clean, particularly in high-use areas such as the cloning and protein purification benches. Autoclave and sterilize lab consumables, including tips, plates, and tubes, to maintain a sterile and efficient work environment. (10%)
